What happened

Lilly announced detailed TRIUMPH-2 results on Sept. 29, 2026. The trial studied retatrutide in adults with obesity and type 2 diabetes. AJMC reports the paper was published in The Lancet with a presentation at the European Association for the Study of Diabetes meeting, dated Sept. 30.

On the efficacy estimand, average weight loss was 12.7% at 4 mg, 19.1% at 9 mg and 20.8% at 12 mg, against 4.0% on placebo. Lilly reports that 10.8%, 30.6% and 34.9% of participants lost 25% or more, against 0.8% on placebo. AJMC adds 23.4% average loss among participants with a baseline BMI of 35 or higher at 12 mg.

A1C fell by an average of 1.4% (4 mg), 1.6% (9 mg) and 1.5% (12 mg), against 0.2% on placebo. Discontinuation for adverse events was 3.8%, 11.6% and 7.7% by dose, against 4.9% on placebo.

Lilly says it plans to submit a biologics license application in the first quarter of 2027.

What remains unknown

  • The Lancet paper itself, including enrollment: it returned an access error. Another summary circulating gives a lower 12 mg average (18.8%); we could not tie it to a source. A treatment-regimen estimand would typically be lower than the efficacy estimand reported here.
  • Long-term safety beyond the 80-week trial.
  • When, or whether, FDA will approve retatrutide.
